A novel flow-injection chemiluminescence-based method has been developed for determination of superoxide dismutase (SOD) activity. An in-vitro superoxide anion generation xanthine/xanthine oxidase stable source was established on line with FIA/CL-detection apparatus, for measuring SOD activity. This method can detect SOD in the linear range of 0.002–2.00 U mL–1 with a detection limit of 0.001 U mL–1. Another method for detection of superoxide anion is based on the luminol–FeCl3 chemiluminescence (CL) reaction. This method was used to evaluate superoxide release and SOD activity in rats treated with the traditional Chinese herb Pulsatilla chinensis, which resulted in high clearance of hepatitis B virus (HBV) after treatment of a hepatitis B patient. Interestingly, we found that treatment with Pulsatilla chinensis can specifically increase superoxide release by liver tissues and, at the same time, slightly increase extracellular SOD (ECSOD) activity in plasma; in particular it can markedly increase MnSOD activity in mitochondria in liver tissue. This work revealed a possible mechanism whereby Pulsatilla chinensis prevents possible infection (for example HBV) by specifically increasing superoxide release in the liver and increasing MnSOD activity to minimize superoxide-mediated toxicity. 相似文献

Peroxynitrite, as a derivative of nitric oxide, is a potent oxidant. It reacts with several biological molecules, makes cellular and tissue damages, and is related with many diseases; therefore, it is of major concern in current medical research works. In this work, a special perm-selective cellulose acetate membrane sampler is used to implement flow injection analysis (FIA)/chemiluminscence (CL)-detection method for the detection of peroxynitrite with Luminol CL-reagent. Optimum detection conditions were established, and the permeability of peroxynitrite through cellulose acetate (CA) membrane, as well as the interference from matrix constituents were studied. The proposed method has the high sensitivity of the CL-detection and the selectivity of perm-selective membrane sampler. The obtained detection limit of 1×10−11 M (without dialysis membrane) and 1×10−10 M (with dialysis membrane), makes it possible to monitor the elusive peroxynitrite in biological samples. The mechanism of luminol CL-emission generated during oxidation by peroxynitrite and the kinetics of peroxynitrite decomposition were also studied using FIA/CL-detection set-up. 相似文献

Hydroxyapatite (HAp) was prepared using a microemulsion route in combination with the pH-shock wave method. The samples as received consisted of amorphous aggregated particles, which had remarkable mesoporosity with a narrow pore size distribution. After being heated at 650 degrees C, the A-type carbonate hydroxyapatite was crystallized at 635 degrees C in particles of similar size (40--120 nm) with no internal porosity. At a higher temperature (900 degrees C) a sintering process took place, resulting in network of a larger particles, consisting of HAp and beta-tricalcium phosphate (beta-TCP). The crystallization of HAp occurs at 635 degrees C with an activation energy of 62.7--72.2 kcalmol(-1). 相似文献

The influence of aspect ratio in three‐dimensional, numerical experiments of separated flows is studied in the case of the backward‐facing step at Reynolds numbers 600, 800, and 950. The computational domain is designed as an actual laboratory experiment. The governing equations are the steady state, isothermal, and incompressible Navier–Stokes equations. The expansion ratio of the computational domain is 1:2. The aspect ratio varies from 1:10 to 1:40. The results of the computations focus on the spanwise variations of the length and the strength of the two eddies along the lower and upper wall. It is concluded that both numerical and laboratory experiments should be designed with an aspect ratio of at least 1:20, if only the accuracy of the position of the detachment and the re‐attachment points matters. If the accuracy of the shear‐stress distributions is also taken into account, then an aspect ratio of at least 1:30 should be chosen. Finally, if the magnitudes of the vortex centers are also considered, then only the aspect ratio of 1:40 qualifies for a realization of two‐dimensional flow conditions in the plane of symmetry. This is contrary to the common practice in the field, at least from the side of laboratory experiments, where an aspect ratio of 1:10 is still considered adequate for this purpose. This report discusses a new approach for the resolution of the fluid-dynamic limit for the Broadwell system of the kinetic theory of gases, appropriate in the case of Riemann, Maxwellian data. Since the formal limiting system is expected to have self-similar solutions, we are motivated to replace the Knudsen number in the Broadwell model so that the resulting model admits self-similar solutions =x/t and then let go to zero. The limiting procedure is justified and the resulting limit is a solution of the Riemann problem for the fluid-dynamic limit equations. A class of Riemann data for which this program can be carried out is exhibited. Furthermore, it is shown that for the Carleman model the complete program can be done successfully for arbitrary Riemann data. 相似文献

Lactobacillus bulgaricus grown on whey was dried by a simple thermal drying method at the range 35–55°C and its efficiency for lactic acid fermentation
of whey was evaluated. Drying of cells in whey suspension in the examined temperature range did not affect significantly their
viability (82–87% survival), indicating a protective effect of whey as both growth and drying medium. The kinetics of fermentation
of whey and mixtures of whey/molasses using the dried culture were comparable to those of non-dried cells, and only low pH
had a detrimental effect on the fermentation ability of the dried cells. Furthermore, dried L. bulgaricus, free or immobilized on casein coagulates, was used as starter for the production of unsalted hard-type cheese. The effects
of the amount of starter culture and the immobilization technique, the evolution of microbial counts, and the sensory properties

Evacuations are massive operations that create heavy travel demand on road networks some of which are experiencing major congestions even with regular traffic demand. Congestion in traffic networks during evacuations, can be eased either by supply or demand management actions. This study focuses on modeling demand management strategies of optimal departure time, optimal destination choice and optimal zone evacuation scheduling (also known as staggered evacuation) under a given fixed evacuation time assumption. The analytical models are developed for a system optimal dynamic traffic assignment problem, so that their characteristics can be studied to produce insights to be used for large-scale solution algorithms. While the first two strategies were represented in a linear programming (LP) model, evacuation zone scheduling problem inevitable included integers and resulted in a mixed integer LP (MILP) one. The dual of the LP produced an optimal assignment principle, and the nature of the MILP formulations revealed clues about more efficient heuristics. The discussed properties of the models are also supported via numerical results from a hypothetical network example. 相似文献

We consider the problem of self-similar zero-viscosity limits for systems ofN conservation laws. First, we give general conditions so that the resulting boundary-value problem admits solutions. The obtained existence theory covers a large class of systems, in particular the class of symmetric hyperbolic systems. Second, we show that if the system is strictly hyperbolic and the Riemann data are sufficiently close, then the resulting family of solutions is of uniformly bounded variation and oscillation. Third, we construct solutions of the Riemann problem via self-similar zero-viscosity limits and study the structure of the emerging solution and the relation of self-similar zero-viscosity limits and shock profiles. The emerging solution consists ofN wave fans separated by constant states. Each wave fan is associated with one of the characteristic fields and consists of a rarefaction, a shock, or an alternating sequence of shocks and rarefactions so that each shock adjacent to a rarefaction on one side is a contact discontinuity on that side. At shocks, the solutions of the self-similar zero-viscosity problem have the internal structure of a traveling wave. 相似文献
